PFAS Mixtures Are Synergistically Toxic to Human Heart Cells

One PFAS compound damages heart cells. Three together do synergistic damage, killing cells faster and destroying the very proteins that make a heart beat.
Worse Together
A 2026 study in Chem Res Toxicol exposed human heart cells (derived from stem cells) to three PFAS compounds: PFHxS, PFOA, and PFDA. Individually, each caused some damage. Combined, the toxicity was synergistic, meaning worse than the sum of individual effects.
After just 5 to 10 days of combined exposure, heart cell viability dropped significantly. After 14 days, the cells started losing their identity as heart cells. Key cardiac markers (NKX2.5, α-actinin, cardiac troponin T) decreased. Vimentin, a marker of scar-forming cells, went up.
Mitochondria Destroyed
Combined PFAS wrecked the heart cells' power supply. Mitochondrial superoxide (toxic free radicals) increased. ATP production dropped. Cellular respiration decreased. The mitochondria were failing, and heart cells were running out of energy.
Protein analysis revealed damage across extracellular matrix organization, cholesterol metabolism, and antioxidant defenses. The entire infrastructure that keeps heart cells alive and functioning was compromised.
Real-World Relevance
Nobody is exposed to just one PFAS. The doses used matched levels found in occupationally exposed workers, but general population levels are climbing toward those ranges. Heart disease is already the #1 killer. PFAS may be making it worse.
